La pícara Justina se adscribe al subgénero narrativo de novela picaresca, inaugurado por el Lazarillo de Tormes en 1554. Formalmente, estamos ante una autobiografía ficticia en la que un personaje cuenta su vida desde muy joven hasta alcanzar la edad adulta. Es un personaje de muy baja condición social, sin apenas principios morales y con tendencia al robo, la mentira y todo tipo de argucias de las que pueda extraer ganancia. La crítica social es fuerte y continua; se denuncia la hipocresía, la avaricia y el egoísmo atroz que recorre la sociedad, sin distinción de clases, posición o sexo.

La gran novedad de esta novela es que la protagonista es mujer. Ya existía el precedente de La lozana andaluza (Venecia, 1528), escrita por el médico Francisco Delicado; es cierto que esta obra se inscribe más en la tradición de La Celestina que en la picaresca. Justina Díez cuenta su vida desde los dieciocho años hasta la madurez avanzada, aunque el relato principal se detiene con su boda, más o menos a los veinte años de edad. La protagonista narra sus engaños y trapacerías para sobrevivir en un mundo hostil, tanto por mujer, como por pobre.

Es una chica espabilada, astuta y un tanto desvergonzada. Pronto comprende que forma parte del eslabón débil de la sociedad, por lo que trata de paliar esta situación explotando su palabrería, su facilidad para las zalemas y su artero modo de embaucar a hombres a quienes gustaba; ella, sin embargo, los desprecia y, a poco que la situación sea propicia, humilla, en público y en privado. En general, los vapulea dialécticamente, los ridiculiza por sus pretensiones amorosas para con ella, o los somete a episodios donde acaban embadurnados de excrementos, o con la bolsa vacía, o las dos cosas a la vez.

El final de la novela coincide con su matrimonio con un hombre viudo de su pueblo, que en el fondo es su protector, Lozano. No es que sea un modelo de marido, pues sabemos que es algo violento, un tanto mujeriego y empedernido tahúr. Se pueden decir muchas cosas de Justina, pero, en efecto, como proclama su nombre, es justa analizando a los demás, y a sí misma, señalando vicios y virtudes sin tapujos.

Como se puede apreciar en la lectura del texto, Justina es viajera, y no por necesidad, sino por gusto. Le encanta recorrer caminos, en general, a lomos de su burra, visitar pueblos y ciudades, acudir a romerías, etc. Vive en Mansilla, pero visita, por este orden, Arenillas (un pueblo, o acaso paraje con ermita --que hoy no existe--, en la provincia de Palencia), León, La Virgen del Camino, Medina de Rioseco (Valladolid) y Mansilla otra vez. En este sentido, el movimiento narrativo es circular, pues el relato comienza y acaba en el mismo punto.

Allí donde llega Justina, tantea la situación y sus posibilidades tanto de divertirse (sobre todo, ridiculizando y riéndose de los demás), como de lucrarse (principalmente, engañando a quien puede para obtener dinero o bienes apetecibles). Pone en marcha sus planes, pues tiene una cabeza despejada y rápida, y, tras la diversión, se retira a otro lugar o vuelve a casa. En su visita a León lo acompaña un muchacho, en oficio de mochilero, llamado Lorenzo; es un briboncete, pícaro en potencia. Este modus operandi se altera algo en su estancia en Medina de Rioseco, a donde va por un pleito, y trabaja de proveedora de hilanderas.
