Die derzeit übliche thermische Behandlung ist im Gegensatz zum Hochdruck mit einigen Nachteilen verbunden. Dazu zählen die Energiekosten, die oft unerwünschten chemischen und physikalischen Veränderung von Inhaltsstoffen sowie die Belastung des Biomaterials durch die Aufheizphase auf die gewünschte Temperatur. Druck hingegen herrscht sofort im gesamten Lebensmittel und ist insgesamt schonender, da er z.B. keine kovalenten Bindungen zu spalten vermag. Um nun Aussagen auf molekularer Basis der Inhaltsstoffe, z.B. Proteine, treffen zu können verwendet man Molekülsimulationen. Numerische Simulation von Biomolekülen in verschiedenen Lösungsmitteln bilden seit vielen Jahren einen Forschungsschwerpunkt. Das liegt zum einen an der Entwicklung neuer Berechnungsalgorithmen und Modelle, zum anderen an der rapiden Zunahme an Rechenkapazität. Eine der ersten molekulardynamischen Simulationen unter Hochdruck führten Kitchen et al. [ ] im Jahr 1992 durch. Sie untersuchten BPTI (bovine pancreatic trypsin inhibitor) in Lösung bei einem Druck von 1000 MPa. Es gilt dabei zu bedenken, dass die damaligen Rechner nicht annähernd die Rechenleistung besaßen wie die heutigen, und damit die Simulationszeiten im Bereich von 100 ps lagen. Sie beobachteten, dass keine druckinduzierte Denaturierung in dieser kurzen Zeitspanne zu erkennen war. Heutige Rechnernetzwerke bringen ein Vielfaches der Leistung damaliger Rechner, sodass Beobachtungszeitspannen von 10-100 ns möglich werden.
