Diplomarbeit aus dem Jahr 2006 im Fachbereich Biologie - Mikrobiologie, Molekularbiologie, Note: 1,0, Christian-Albrechts-Universität Kiel (Biochemie), Veranstaltung: Biochemie/Molekularbiologie, Sprache: Deutsch, Abstract: Das Protein Tumornekrosefaktor-alpha (TNF-α) wurde zum ersten Mal 1984 von Pennica et al. aus einer Monozyten-Zelllinie isoliert und sequenziert. 1985 beschrieb Old, dass durch TNF-α in mit Lipopolysacchariden (LPS), dem Endotoxin gram-negativer Bakterien, kontaminiertem Tumorgewebe Nekrose und zum Teil die vollständige Regression der Tumoren im Tiermodell ausgelöst wurde. Man hielt TNF-α zunächst für ein bakterielles Produkt. Erst später entdeckten Granger et al., 1969; Carswell et al., 1975; Fransen et al., 1986, dass Antigene (hauptsächlich gegen LPS) in den Patienten für die Sekretion des körpereigenen Zytokins, TNF α, verantwortlich waren, welches u.a. in Tumorzellen Lyse und Zelltod auslösen kann. Als therapeutisches Mittel zur Anti-Tumortherapie eignet sich TNF-α aufgrund seiner hohen Zytotoxizität jedoch nicht. Das TNF-α-Gen ist auf 6p21.3 lokalisiert und besteht aus 4 Exons, welche für einen Precursor aus 230 Aminosäuren und einem Molekulargewicht von 26kDa kodieren. TNF-α wird zunächst als transmembranes Protein mTNF (engl. membrane-bound TNF-α) exprimiert und durch die Metalloprotease TACE (engl. TNF-α converting enzyme) proteolytisch gespalten. Das gesplicte Protein besteht aus 157 Aminosäuren mit einem Molekulargewicht von 17kDa und wird als lösliche Form sTNF (engl. soluble TNF-α) freigesetzt, welche Homotrimere bildet. TNF-α und das Lymphokin Lymphotoxin β (TNF-β), welches aus Lymphozyten stammt, haben ähnliche biologische Aktivitäten und zeigen eine Aminosäure-Übereinstimmung von 30%. Die Wirkung von TNF-α ist proinflammatorisch und nimmt bei der Abwehrreaktion des Körpers eine wichtige Rolle ein.
