Der umfangreiche Band enthält 425 Briefe, davon 112 Briefe zwischen Marx und Engels, 37 von ihnen an dritte Personen gerichtete und 278 an sie adressierte Briefe. Von den Briefen Dritter werden 221, also die Mehrzahl der in diesem Band dargebotenen Dokumente, erstmals oder (zehn Briefe) erstmals in der Originalsprache bzw. vollständig veröffentlicht.

Die Briefe zeichnen ein intensives Bild der Reaktion von Marx und Engels auf die zeitgenössischen politischen und wirtschaftlichen Entwicklungen. Große Aufmerksamkeit schenken sie dem Amerikanischen Bürgerkrieg. Vor allem 1862, als Marx in seinen Korrespondenzen der Analyse der Vorgänge in den USA Vorrang einräumte, verständigen sie sich darüber laufend. In Europa galt ihr Interesse dem Heeres- und Verfassungskonflikt in Preußen sowie dem Krieg um Schleswig und Holstein 1864. Vom polnischen Januaraufstand 1863 erwarteten sie kurzzeitig gar den Beginn einer neuen europäischen Revolution.

Die Korrespondenz zeigt ihre leidenschaftliche Parteinahme für den polnischen Freiheitskampf und gibt Aufschluss über die Broschüre, die Marx und Engels hierzu abfassen wollten. Insgesamt dokumentieren die Briefe oft zugespitzter als in gleichzeitigen Veröffentlichungen ihre politischen Erwägungen, die erörterten Meinungsverschiedenheiten und auch Gedanken, die in ihren Publikationen keinen Niederschlag gefunden haben. Zudem geben sie Einblick in die Arbeit von Marx, hauptsächlich an dem ökonomischen Manuskript 1861-1863, eine wichtige Phase der Entstehung seiner ökonomischen Theorie.

Bei einem gewichtigen Teil der Korrespondenz zwischen Marx und Engels handelt es sich um intime Privatbriefe, die in dieser Dichte selten überliefert sind. So fällt in diese Zeit die Einstellung der Briefkontakte zu Ferdinand Lassalle im Jahre 1863 - fortan sollten Marx und Engels nur noch über ihn, aber nicht mehr mit ihm kommunizieren - sowie das kurzzeitige Zerwürfnis zwischen Marx und Engels. Grund war Marx' unsensible Reaktion auf den Tod von Engels' Lebensgefährtin Mary Burns, in Anbetracht seiner eigenen finanziellen Misere. Es ist dies wohl der einzige dokumentierte ernsthafte Konflikt zwischen beiden.
