Seu maior trabalho é o enorme conjunto de cinco volumes de livros Adversus Haereses, ou Contra as heresias, livros estes que apresentamos aqui em um só volume. Temos aqui uma refutação das doutrinas do gnosticismo. Além de sua proximidade com João e os Pais Apostólicos, os escritos de Irineu são todos completamente universais. Os movimentos gnósticos heréticos levaram Irineu a desenvolver a teologia da Igreja e a cristologia, ou um entendimento de exatamente quem é Cristo. Irineu desenvolveu a idéia da necessidade de uma expiação e redenção corporais através da humanidade sagrada de Jesus. Para entender isso melhor, devemos primeiro olhar para os falsos ensinamentos do gnosticismo.As seitas gnósticas enfatizavam um conhecimento secreto, pseudo-místico, que tinha que ser adquirido para a salvação, e geralmente reservado apenas para os poucos que eram considerados espiritualmente dignos. Como tal, o gnosticismo tornou-se associado ao elitismo. A maioria dos mitos gnósticos, baseando-se fortemente na filosofia pagã grega, ensinava que as coisas do mundo eram criadas por um semi-deus perverso, Demiurgo, e, portanto, o mal. O universo material maligno está então em desacordo com a bondade do Criador Supremo e o mundo espiritual. O gnosticismo desceu para uma forma de dualismo, onde o corpo e toda a matéria são maus, e tudo o que é espiritual é bom. O mundo, e tudo o que está nele, deve ser rejeitado.
