Inhaltsangabe: Einleitung: Die vorliegende Diplomarbeit beschäftigt sich mit der Auswertung von Patch-Clamp-Messungen. Die Patch-Clamp-Technik, mit deren Hilfe der Strom durch einzelne Ionenkanäle einer biologischen Membran gemessen werden kann, ermöglicht die Erforschung von Schaltprozessen der Transportmoleküle. Die Analyse der mit der Patch-Clamp-Technik gemessenen Kanalströme geht davon aus, dass das Verhalten der Kanäle durch Markov-Modelle beschrieben werden kann. Markov-Modelle haben eine endliche Anzahl diskreter Zustände, zwischen denen das Transportmolekül wechseln kann. Die Übergangswahrscheinlichkeiten zwischen den verschiedenen Zuständen verändern sich nicht mit der Zeit und hängen insbesondere nicht davon ab, wie lange das Transportmolekül in einem Zustand verweilt hat und in welchem Zustand es vorher war. Dieser einfache Aufbau der Markov-Modelle führt dazu, dass sie sich gut mathematisch behandeln lassen. Ziel der Arbeit ist es, das Zeitauflösungsvermögen für die Bestimmung der Übergangsraten zwischen den verschiedenen Zuständen des Markov-Modells zu verbessern. Der konkrete Ansatzpunkt liegt darin, im "Direkten Zeitreihenfit" den bis jetzt vernachlässigten Einfluss des Tiefpassfilters auf die Verteilung der Stromwerte zu berücksichtigen. Im "Direkten Zeitreihenfit", einem Verfahren, das ursprünglich für die Spracherkennung entwickelt wurde, wird mit einem Prädiktionsalgorithmus ein Wert für den nächstfolgenden Stromwert vorhergesagt und dieser vorhergesagte Wert mit dem tatsächlichen Messwert verglichen. Zusammenfassung: Ergebnis der Arbeit ist es, dass durch geeignete Wahl der Parameter das Zeitauflösungsvermögen gegenüber vorhandenen Auswerteverfahren um den Faktor 10 gesteigert werden kann. Es können damit Übergangsraten bis zum Zehnfachen der Abknickfrequenz des Tiefpassfilters erkannt werden.
